I used to use ECO mode all the time until one day I was waiting for an opening to make a left turn and tried to scoot across when an opening came. I pressed the gas pedal and it took about two full seconds before it responded. I almost got hit! Now, I mostly use SPORT for the responsive throttle. I still use ECO on the highway.

I’ve never picked up on the engine sound enhancement (ESE) in Sport. Is that actually a thing in the TRD Pro?
Oh yeah. Try it out and you can hear the difference pumped into the cabin.

Eco. But what the hell is comfort mode???
According to what I found on the internet:

Comfort mode in a Toyota vehicle adjusts the suspension to provide a softer ride and lightens the steering feedback, making driving more relaxed and enjoyable. It is designed to enhance comfort, especially on smooth roads or highways.

Either normal or tow/haul. Tow/haul does the same as Sport without the annoying noise.

So far I've been driving exclusively in ECO. I tried Sport a few times and really liked the pep but didn't care for the artificial sound. Granted the turbo 4 cylinder has a lame sound without it but it just was weird sounding. Might do Normal in heavy holiday traffic when stuck on 476 with bumper to bumper traffic so that it stays closer to the person in front of me.

I use Normal mostly and sometimes Sport depending on how I feel that day.

I run normal. Two reasons: I hate the artificial noise in Sport. And I never bother to change out of the default normal at start to get into Eco. MPG impacts seem all over the map so I go with simple and unamplified.
Also... only Limited and Platinum have the Adaptive Variable Suspension system and the Comfort Sport S/S+ and Custom modes that come with that. Relatively small proportion of 6G4Rs - IF your question is focused on hearing about those modes....
Normal does not seem to be the "default" in my SR5. I set to Eco and it stays in that mode unless I change to normal or sports. Maybe its just the Sports mode that you cannot leave in that mode?

I like to use Eco. Normal is okay, but not peppy enough to justify the decreased MPG. Sport mode is fun for the mountains, and helpful to merge onto the highway or pass, but I set the mode back to Eco as soon as I am cruising again.

Rotating the mode dial makes me feel like I'm controlling a fighter jet or something, so admittedly I find excuses to use it.

Holy Crap - I just tested this and you seem to be right. When in Eco it stays there after engine off and then starting again later, but when in Sport it reverts to Normal at next start**. At least that is what happened to me today. (And I reminded myself how much I hate the artificial engine noise in sport:>) Might set in Eco and see how it feels. I've got enough detailed milage data to do a solid analytic comparison with Normal.

**Normally I take the time to review the manual on such details but I'm going to assume that either this is not clearly stated or I simply glossed over it. Real evidentiary experience is better than trying to interpret the manual anyway.

I like the topic. I have not been able to get beyond 20 mpg with around town (20%) the rest at 40 or highway speed. As I recall at 60-65 the MPG dropped below 20.
The setting is always normal. I am around 800 miles.

However the system went into fault when the air/fuel sensor crapped out a couple weeks ago. (still waiting for the part).
This sensor affects the mix, wonder if it was running rich?
